You can use a smartbomb to destroy missiles, but you need to time it correctly, which is tricky as you need to time the smartbomb delay to hit the missile when it's in range. I've not heard of defenders or smart bombs being able to destroy rocket...

Defenders need time to lock, then fly to and hit enemy missiles. I find they're pretty much useless at less than 20km. If you just want to pack them to stop enemy missiles, then I'd advise sticking them in a rocket launcher, which has a faster ROF...
